Dr. Mosoni's presentation delved into how Eggerthella lenta metabolizes flavan-3-ols—polyphenols known for their cardiovascular benefits—into bioactive compounds in the gut. By identifying ten key genes responsible for this metabolic process through a transcriptomic approach, Dr. Mosoni's work has revealed that 27% of individuals lack the ability to convert these polyphenols, raising important questions about cardiovascular risk factors.

About Dr. Mosoni

Dr. Mosoni is a microbiologist in MEDIS (Microbiology, Digestive Environment & Health) laboratory at INRAE. Her research activities are currently focusing on the metabolism/degradation of dietary fibers and polyphenols by the human gut microbiota in order to better understand the effects of these plant dietary components on health through the action of the microbiota. The approaches used involve the isolation of anaerobic microorganisms contributing to these metabolic pathways and characterize their function (genes, enzymes, metabolites) and role in health. Her aim is to discover new generation pre- pro- syn-biotics, or effective bioactive molecules (postbiotics) to be used in preventive nutritional strategies.

Pascale Mosoni of INRAE has won the Best Short Oral Presentation Award at Polyphenols Applications 2024 for her research on “The Molecular Basis of the Degradation of Flavan-3-ols by the Human Gut Bacterium Eggerthella Lenta.”

Dr. Laure Poquet of Nestlé Research has been awarded the Best Poster Presentation (1) at Polyphenols Applications 2024 for her outstanding research on oleuropein metabolites.

Her poster, “Oleuropein Metabolites Are Biologically Active as Mitochondrial Calcium Activators at the Concentration Measured in Human Plasma,” highlighted the bioactivity of these metabolites derived from olive leaf extracts.

The study demonstrated that, at concentrations measurable in human plasma, these metabolites can activate mitochondrial calcium, leading to enhanced energy metabolism in muscle cells.

A new study was performed by Nestlé scientists to understand how the polyphenols from olive leaf extract can boost muscle energy. The main natural polyphenolic compound of this extract is oleuropein. Its metabolites circulating in plasma after intake were detected and quantified using a validated LC-MS/MS method. Their potential biological activities as mitochondrial activators were tested in vitro, at the circulating concentrations. The results showed that, although the level of oleuropein was low, its metabolites are biologically active in skeletal muscle cells and reach a total concentration in human blood sufficient to activate mitochondrial energy metabolism via mitochondrial Ca2+ activation.

This achievement marks a significant step in understanding the role of oleuropein in improving muscle function and supporting healthy aging.
Presentation Summary

Cellular senescence is characterized by a stable cell proliferation arrest and the establishment of a secretory phenotype in normal cells in response to intrinsic and extrinsic stimuli.

Cellular senescence is associated with chronological aging and a response to environmental stressors.

Expanded knowledge about the role of senescence in the physiological context and disease processes and development of drugs that can counteract the harmful effects of cellular senescence as a therapeutic target.

Therefore, senotherapeutics can be classified as senolytics drugs that selectively eliminate senescent cells or senostatics (senomorphic) that inhibit, reverse, or prevent aspects associated with cellular senescence.

The most studied option combines dasatinib and quercetin, a natural polyphenol that targets BCL-2 and PI3K/AKT signaling pathways.

The various pharmacological mechanisms described for polyphenols suggest that others may have senotherapeutic effects, which has been the focus of our recent work.

Presentation Topic: Coevolutionary Role of Host and Gut Microbiota in Polyphenol Health Effects: Metabotypes and Precision Health.

Presentation Summary

Individuals can be grouped into different metabotypes (metabolic types) based on their gut microbiota's ability to metabolize certain polyphenols, resulting in unique metabolites with varying molecular structures and biological activities. Thus, the effects of certain polyphenols will greatly depend on each individual's gut microbiota. By understanding the microbial ecologies responsible for these metabolic pathways, "precision-health" strategies could be developed to standardize the effects of polyphenols.

About Juan Carlos Espín

Juan Carlos Espín is is a full Research Professor at CEBAS-CSIC (Murcia, Spain). For the past 24 years, his main lines of research have focused on: i) the metabolism and biodistribution of polyphenols in humans, identifying the final responsible actors for biological effects; ii) the use of exosomes to deliver and enhance the biological activity of polyphenols and derived metabolites in systemic tissues; and iii) understanding why some polyphenols do not exert the same effect in all individuals. His research suggests that specific gut microbial ecologies play crucial roles in mediating the observed effects within the context of "precision health".

Juan Carlos has been a "highly-cited" researcher consecutively from 2018 to the present (only 0.1% of researchers worldwide are "highly-cited"). As of March 2024 (according to Scopus), he has an H-index of 85, with 230 JCR publications totaling approximately 23,000 citations, and has led the second highest-earning patent in the history of the CSIC.

She will present the Novel Development on the Potential Application of Pyranoanthocyanin as Colorants.

Presentation Summary

Pyranoanthocyanins are derived from anthocyanins and are key contributors to the extra long stability of red wine color.

Yet, their limited stability limits their application in foods.

Different strategies were developed by Prof. Gusti and her team to produce a wide variety of pyranoanthocyanins from an array of different anthocyanin precursors, and in combination with different cofactors.

These pyranoanthocyanins show superior stability to factors such as pH, bleaching agents and heat.

They also present new challenges such as their decreased solubility in water and the low efficiency of production.

Different strategies to increase the yields of pyranoanthocyanin production from different sources will be discussed, as well as the role of their chemical structure on pyranoanthocyanin color, solubility, stability and potential for food applications.

The heterogenous class of polyphenols exhibit surprising and interesting antiviral effects.

The COVID-19 pandemic has also inspired research on polyphenols and their health promoting properties.

Current research including our own findings show that prenylated flavonoids (e.g. 6-prenylnaringenin) inhibit important viral enzymes of SARS-CoV-2.

Also other polyphenols (like quercetin and curcumin) exert antiviral effects on SARS-CoV-2.

Polyphenols seem to be promising natural drug candidates for other human viral diseases (e.g. hepatitis, influenca).

Presentation Summary

2-ene reductases are NADH-dependent enzymes that specifically reduce double bonds conjugated with an aromatic ring.

Plant-associated lactobacilli possess multiple homologues of HcrB, Par1, and other related enzymes from different bacteria.

These enzymes are involved in the conversion of various phenolic compounds, indicating a diverse enzymatic capability within lactobacilli.

The diversity of 2-ene reductases in lactobacilli suggests their role in metabolizing and modifying phenolic compounds.

This enzymatic diversity may have implications for the metabolism of dietary phenolics and potentially contribute to the functional properties of lactobacilli in various ecological niches.

The Polyphenols Applications 2023 committee acknowledges Dr. Charareh Pourzand from the University of Bath, UK, for her exemplary achievement in securing the Best Short Oral Presentation Award.

In her presentation titled "Hydroxycinnamic Acid Derivatives as Dual-Action Antioxidants and Iron Chelators for Shielding Skin Cells from UVA Photodamage", Dr. Pourzand provided a comprehensive overview of her research findings. Among the multiple hydroxycinnamic acid derivatives studied, trans-cinnamic acid (TCA) emerged as particularly potent in its photoprotective efficacy against UVA in skin cells. This was attributed to its pronounced capability to reduce both basal and UVA-induced increase in intracellular labile iron levels. Dr. Pourzand elaborated on the dual mechanistic role of hydroxycinnamic derivatives, with a notable emphasis on TCA, in counteracting UVA-induced damage in skin cells.

In general, Dr. Pourzand's Research on UVA photobiology specially in relation to skin photoaging has revealed that UVA component of sunlight has a dual damaging effect on skin cells. In one hand it generates harmful reactive oxygen species (ROS) and on the other hand it promotes the release of harmful free iron that acts as a catalyst to exacerbate the damaging effect of ROS, leading to irreversible skin photodamage in the form of wrinkling, sagging, etc .

Dr. Pourzand stated: "To counteract this dual effect of UVA, we are in process of identifying natural compounds based on polyphenols that can not only neutralise the UVA-induced ROS but also mop the free iron released by UVA. Since the properties and the concentration of such polyphenols can significantly change in different seasons or locations, our future research is concentrated on designing 'bio-inspired' molecules that are more stable than their natural counterparts but still maintain their dual functions. Such bifunctional compounds can then be added as photo-protectants into sunscreen formulations for a more robust protection against UVA photodamge exerted by sunlight".

Such advancements in the field of skin photoprotection are of immense importance, and the Polyphenols Applications 2023 team, along with the broader scientific community, recognizes and appreciates Dr. Pourzand's significant contribution.

Dr. Zimmermann and his colleagues developed a new natural polyphenol supplement that strongly chelates iron in vitro and assessed its effect on non-heme iron absorption in patients with hereditary hemochromatosis (HH). The polyphenol supplement reduced iron absorption by ~ 40% from meals and test drinks.

Their results indicate that when taken with meals, this natural polyphenol supplement can decrease dietary iron absorption, and might thereby reduce toxicity of body iron accumulation in patients with HH.
